AN ENZYMATIC METHOD FOR THE DETERMINATION OF UREA IN AQUEOUS SAMPLES USING CRUDE UREASE FROM Citrullus lanatus SEEDS

This paper presents an enzyme-based method for the determination of urea in aqueous samples using crude urease from Citrullus lanatus seeds. The is based on enzymatic hydrolysis Urea into ammonia (NH4) and carbon dioxide (CO2) by urease. Ammonia formation determined spectrophotometrically after nesslerization then quantitatively related to concentration via a calibration curve. Reaction conditions assay were investigated optimized. Optimum temperature was found be 50 oC, optimum pH 8.0 reaction time 10 minutes. Calibration curve linear (R2 =0.987) within range 0.5 – mg/L. Limits detection quantification (LOD LOQ) 0.08 0.13 mg/L respectively. Application surface water revealed good precision (1.45 3.28 % RSD) accuracy (84 92% recovery). presented simple has potential practical tool monitoring environmental samples.
